AMPK

AMPK (AMP-activated protein kinase) is the cell's master energy sensor. It activates when cellular energy is low — during exercise, fasting, or caloric restriction — triggering autophagy, fat burning, and mitochondrial biogenesis. AMPK activation is one of the most consistent signatures of longevity interventions.

AMPK senses the cellular ratio of AMP to ATP. When ATP is abundant, AMPK is quiet. When ATP is depleted and AMP rises (during exercise, fasting, or cold exposure), AMPK is activated through phosphorylation by upstream kinases including LKB1 and CaMKK2. Once active, AMPK functions as a master switch: it turns on energy-producing catabolic processes and turns off energy-consuming anabolic processes.

Downstream, AMPK activation: suppresses mTOR (shifting cells from growth mode to maintenance mode), activates autophagy, increases fat oxidation in muscle and liver, translocates GLUT4 to the cell membrane (improving glucose uptake independent of insulin), activates PGC-1α (driving mitochondrial biogenesis), and inhibits fatty acid synthesis. This coordinated response is remarkably similar to the signature of caloric restriction, the most reproducible longevity intervention in animal research.

The AMPK/mTOR axis is central to the biology of aging. In youth, these pathways cycle appropriately — mTOR dominates during growth and feeding, AMPK during exercise and fasting. With age, this rhythm flattens: mTOR tends toward chronic activation, AMPK toward chronic suppression, producing the hallmarks of aging (impaired autophagy, mitochondrial dysfunction, accumulated cellular damage).

Natural AMPK activators include: exercise (the most potent), fasting, cold exposure, berberine (a plant alkaloid with evidence for AMPK activation and glucose control), quercetin, resveratrol, and green tea EGCG. For women in perimenopause, restoring AMPK cycling through regular fasting windows and Zone 2 training is one of the highest-leverage interventions available.
